How to Store Liquid Culture Syringes So They Stay Viable

How to store liquid culture is simple: in the fridge at 2 to 8 °C, upright, in the dark and in the original packaging. Mondo’s guidance is that a liquid culture syringe stays viable for 6 to 8 months under those conditions, but a culture used in the first weeks will start faster than one that has been waiting for half a year.

Why does liquid culture need the fridge?

Liquid culture needs the fridge because it contains living mycelium in a nutrient broth. At room temperature the mycelium keeps growing, uses up the nutrients and eventually turns into a dense mat that is hard to push through a needle. Cold slows the mycelium down without killing it.

The same broth would feed bacteria or mold if any got in. Cold slows those down too, which is the second reason the fridge matters.

How should you store a liquid culture syringe?

1
Fridge, not freezer. Keep it at 2 to 8 °C. Freezing forms ice crystals that damage the mycelium.
2
Upright, needle cap closed. Keep the protective cap on the syringe tip and store the needle separately in its own sterile pack.
3
Dark. The fridge is dark most of the time. Do not store syringes in a clear box on the counter.
4
Away from food. Put the syringes in a clean, closed container on a shelf of their own, away from raw meat, cheese and open fruit.
5
Stable. The back of a middle shelf is more stable than the door, which warms up every time the fridge is opened.

Write the date on it

Write the delivery date on the pack. With more than one strain in the fridge it is easy to lose track, and the date tells you which syringe to use first.

How long does a liquid culture syringe last?

A Mondo liquid culture syringe lasts 6 to 8 months in the fridge. That is the outer limit for a reliable result, not the moment it suddenly stops working. Vitality declines gradually: a culture at month seven usually still grows, just more slowly at the start.

For comparison, a spore syringe keeps 8 to 12 months, because dormant spores use almost nothing while they wait. If you want to keep genetics for years, a spore vial is the format made for that, and our liquid culture vs spore syringe comparison explains the other differences.

What should you check before using a stored culture?

Check three things before you inject a culture that has been stored:

  • Clarity. The liquid itself should be clear, with visible white clumps or strands of mycelium. Cloudy liquid points to bacteria.
  • Color. The mycelium should be white to off-white. Green, black, pink or orange specks are a reason to stop.
  • Only if in doubt, smell. A clean-looking syringe does not need opening. If the liquid looks off, a sour or rotten smell at the tip confirms contamination.

Let the syringe reach room temperature for about an hour before injecting, and shake it gently so the mycelium is spread through the liquid. Our guide to grow kit contamination shows what the common molds look like.

Which storage mistakes ruin a liquid culture?

The most common mistakes have little to do with the fridge itself:

  • Leaving the parcel in a warm car or hallway. A day at 30 °C can do more damage than months in the fridge. Unpack and refrigerate on the day of delivery.
  • Using part of the syringe and keeping the rest. Once a needle has been attached and used, the syringe is no longer sterile. Mondo’s manual advises injecting the full 20 ml into one substrate.
  • Storing it in the freezer to make it last longer. It does not; it shortens the life of the culture.
  • Repeatedly taking it out to look at it. Every warm-up wakes the mycelium a little. Check it once, before use.

Can you use a liquid culture past 8 months?

You can try, but you take the risk yourself. An old culture may still colonize, but it starts slower, and a slow start gives contaminants more chance. If you have a choice, use the oldest syringe first and plan your orders so you inoculate within a few weeks of delivery.

Questions about storing liquid culture

A few days at normal room temperature is usually fine, for example during delivery. Refrigerate it as soon as you can and avoid heat above 25 °C.

Mycelium tends to grow into clumps in a still liquid. Shake the syringe gently before use. If the clump will not pass through the needle, swirl it longer and let it rest at room temperature first.

Very slowly. That is why a culture that has been stored for months can look denser than when it arrived.

No. A used or exposed needle is no longer sterile. Inject the full syringe in one go.

Yes: fridge, 2 to 8 °C, dark and never frozen. Spore syringes keep longer, 8 to 12 months.
